How to Make Cell Culture Media for C2C12 and Satellite Cells

Differentiation Media for C2C12 cells

  • 200 ml DMEM
  • 4 ml horse serum (2%)
  • 3 ml penicillin/ streptomycin
  1. Use a sterile vacuum filter (found on bench)
  2. In the hood, place all ingredients for the media in the top half of the filter
  3. Attach the vacuum filter hose to increase filtration
  4. Detach hose, label new bottle (name/date/ recipe)
  5. Store in fridge

Proliferation Media for C2C12 and Satellite cells

  • 250 ml DMEM
  • 50 ml FBS (fetal bovine serum- includes growth factors)
  • 3 ml penicillin/streptomycin
  1. Use a sterile vacuum filter (found on bench)
  2. In the hood, place all ingredients for the media in the top half of the filter
  3. Attach the vacuum filter hose to increase filtration
  4. Detach hose, label new bottle (name/date/ recipe)
  5. Store in fridge

Proliferation Media for Satellite cells

  • 250 ml Harms
  • 50 ml FBS (fetal bovine serum- includes growth factors)
  • 3 ml penicillin/ streptomycin
  1. Use a sterile vacuum filter (found on bench)
  2. In the hood, place all ingredients for the media in the top half of the filter
  3. Attach the vacuum filter hose to increase filtration
  4. Detach hose, label new bottle (name/date/ recipe)
  5. Store in fridge
