Creating a Lower Buffer Solution

Making 1.5M Lower Buffer Solution

  • 500ml total to make

Materials

  • 91g of tris base (into a beaker with a stir bar)
  • 350 ml of milli Q (from the tap not reservoir) measure in a graduated cylinder

Method

  1. Use pH meter and temp sensor (clean with DI water)
  2. Place pH electrode into beaker after calibrating
  3. First pH will be around 11 (tris base is very basic)
  4. Add the temp sensor meter
  5. Hydrochloric acid (HCL) is used to set pH (be careful with the bottle it is coated, kept under the fume hood in the hallway)
  6. Use dropper to take up the HCL (tip bottle at 45 degrees) and check pH till 8.8, add HCL slowly as drops
  7. After pH is reached take on to the weighing bench (clean pH and temp sensor)
  8. Add 2g of 0.04% SDS (careful is powdery) dissolve on stir bar
  • For the 1 M buffer: 700 ml of milli Q and 121g of tris base. pH is 7.4
